PC1002 2018 Week 5 Renal Practical Report Full Write Up Instructions: 50 marks = 5% of Subject Assessment (Refer also to your feedback from SP1 PC1001 Skeletal Muscle Report) Write must be in your LAB BOOK and submitted on Thursday 6th September 2018 (Week 7, same day as Powerpoint Group Presentations) Your report should have a pre-lab flow chart, title, aim, introduction, materials & methods, results, discussion, conclusion, references and appendix (if required) as per your laboratory report in PC1001 last semester. More guidance is provided in the separate generic guide document. Pre-Lab Flow Chart: Signed on the day by academic staff, a basic description of how the experiment will be conducted. Can be a -1 page before the remaining items below. Title: Short but informative. Aim(s): Short but informative. Introduction: As there is considerable information relating to this practical, try to keep your introduction brief but informative (approximately 2 pages), good quality figures (referenced) are encouraged. Your introduction should clearly and concisely describe the function of the kidneys; water and electrolyte balance, and briefly discuss the characteristics of normal urine. Remember to start with the big picture (not too big) and narrow to the focus of the experiments. Materials and Methods: can refer to the laboratory notes but any deviations from these methods should be noted. Photographs of equipment / methods are allowed, but must NOT identify students. Results Part 1. 4 graphs or 2 graphs (if you can show a water line and saline line on one graph). water/urine flow rate vs. time saline/urine flow rate vs. time water/urine osmolality vs. time saline/urine osmolality vs. time On each osmolality graph show the morning sample (it will not have a corresponding time value, so plot against time = 0 minutes). Provide a brief written description of each graph. A table of the results summarized in the graphs should appear in the appendix i.e. your raw data collected during the prac (makes sure it is neatly written or typed). Part II. You need to put in the tables with appropriate descriptions in the Results. Discussion Part I. Water diuresis Make sure you have answered the following questions in the discussion (in question and answer format e.g. Water Treatment 1. The consumption of water causes .. The questions does not have to appear in the report, as long as it is clear that you are answering each question (use headings, #s or paragraph breaks). Water treatment 1. How does the consumption of water affect the ECF volume and ECF osmolarity? 2. How did the resulting change in ECF osmolarity affect ADH release? (include the receptors involved in detecting the alteration and where the hormone is released from, and whether there was an increase or decrease in hormone level) 3. What are the physiological actions of ADH? 4. How did treatment with water affect urine flow rate and urine osmolarity? 5. The change in ECF osmolarity is also detected by macula densa cells. What effect does this change have on the amount of aldosterone that is released? 6. What is the function of aldosterone on the distal convoluted tubules and collecting duct? Saline treatment 1. How did the treatment with saline affect the ECF volume and ECF osmolarity? 2. How did the resulting change in ECF volume affect the amount of ANP released? 3. How did treatment with saline affect urine flow rate and urine osmolarity? 4. What are the physiological actions of ANP? Morning Sample 1. How did the osmolarity of the morning sample differ from the other samples collected from subject 1 (after drinking water)? 2. What causes the morning urine sample to be different from other samples collected during the day? i.e. think about how the levels of ADH and Aldosterone would change during the night. Part II. Urinalysis Compare the Unknowns with the controls. What is wrong with the people with the Unknown samples Why? References: 5-10 good quality references are required for this report. These should be from at least 3 sources (eg: textbooks, journal articles and QUALITY website sources). In text referencing is required (especially in introduction and discussion).
